Read the trial population first
SURMOUNT-1, published in 2022, randomized 2,539 adults with obesity or overweight and a weight-related complication. People with diabetes were excluded. The trial evaluated tirzepatide against placebo over 72 weeks, including an escalation period.
Those details define the scope of the evidence. Findings in that population should not automatically be applied to people with different conditions or to a different formulation. Even a large randomized trial answers a bounded question.
Separate the mechanism from the outcome
A receptor mechanism explains a route through which a compound can act. A trial measures whether relevant outcomes changed under the conditions tested. Neither can replace the other: a plausible mechanism needs outcome data, and an outcome should be interpreted alongside tolerability and study methods.
When reading a report, record both the primary result and how investigators handled participants who stopped treatment. The analysis population can affect the interpretation of an average change.
Compare studies carefully
A comparison with semaglutide or retatrutide should identify whether it uses a direct comparative trial or separate publications. Differences in study length, baseline characteristics, and statistical analysis make a simple ranking by percentage misleading.
